I am not clear about your data structure, If your data structure is like Matrix and want to calculate specific columns, you could try to use filter in measure like below
measure= calculate(sum([value]), filter(table, [columnname]="a1"))+calculate(sum([column]), filter(table, [columnname]="a2"))
If your structure is like table, you could calculate column directly
measure1=sum([a1])+sum([a2])
