dataset from pbi services

Hi, I'm new in PowerBI, and have a big question.

 

If I upload a dataset from an Excel file, when I try to connect with PowerBi Desktop, I can't make columns or use Query Manager, Why?

My data is generated from PowerApps and saved in OneDrive as Excel file, if I upload on pbi services as dataset, I can refresh the dataset in Onedrive Mode, and its fantastic, but I can't make columns on pbi desktop and the software turns very limited. I need columns for calculate minutes of process from, [finish hour - start hour]. Otherwise, can I do this operation with a measure? If the answer is yes, How?

@lonerwalker once you have live connection to power bi dataset, you cannot make any change to the model, to do so, you have to change your original data model that you published in power bi

in that case, can I do relationships on powerpivot, as dataset. powerbi recognize them?
