I was wondering when you would upload a workbook (Excel) to Power BI Service, if you could import its data into the Power BI Desktop report. I seem to be unable to do this.

Sounds like you have to start looking into Datawarehousing solutions to optimally use PowerBI.
The difference between "Get Data" in power bi service and power bi desktop is that in power bi service, you have two choices as below:

It is possible.
The first option in his screenshot will create a dateset for you.
but modeling your data wont be possible anymore.

The report it has to be tied to is a general KPI report used by many departments. It also does an import from other sources and modifies them in order to be shown in the dashboards properly. Yes, it become today to big for Service and reached to 10 GB limit :(.
