I have something I want to do, but I don't know how to do it, so please help me.
For example, when User in image ① is set as a unique value and data is expressed in a table, the total value will be 4 as shown in attached image ②.
Why is it 4 instead of 5?
Also, I would like to know how to set the total value to 5 correctly.

By only seeing the screenshot, I think one of the user of ja and one of the user of en is the same, so that is why total unique count of all user is four, and I think this is correct.
However, if your business case confirms that total has to be five, because simply 3 plus 2 is 5, and If you want to show total as 5, please try to create a measure like below.
Please check the below picture and the attached pibx fie. I tried to create a sample pbix file like below.
fix version distinct users count measure: =
SUMX ( VALUES ( Data[Locale] ), CALCULATE ( DISTINCTCOUNT ( Data[User] ) ) )
