As tested, i'm afraid it doesn't support to sort column header ascending or descending.
You could add your idea in the Idea forum to make this feature possible in the following version.

Why do you need a slicer for that? If you click on the Date column (the title) in the visual itself, you should be able to toggle between sorting in an ascending/descending order.
The users are requesting this feature. The dates are listed accross the top of my matrix and I don't see an option to toggle ascending/descending.

You are correct. In a matrix visual, the option to sort date does not appear when that field is placed in the column labels section. The moment you place the Date field in the row labels section, that sorting option lights up and see in the image below. I do not know how to solve this.
