This is Matrix. I have columns: LOB ,Year ( values are 2019 and 2020) and Cost. I need a difference between the cost for 2019 and 2020 in the difference column.

Try pivoting the year column in the query editor.
Create a measure and apply in table visual.
Difference = MAX('Table'[2020])-MAX('Table'[2019])

@Anonymous not sure how your data looks like, you can try something like this
Difference =
VAR __prevYear = MAX ( Table[Year] ) - 1
VAR __prevYearSales = CALCULATE ( SUM ( Table[Sales] ), Table[Year] = __prevYear )
RETURN
SUM ( Table[Sales] ) - __prevYearSales
