So right now I have a data table with a bunch of different time stamps. I then used the datediff function to calculate the time difference between certain columns. Now I want to calculate the total time by summing them up, but I want to slice it so I can add and remove columns using a slicer. Is this possible?

Could you please share sample data of your table and post expected result here? In Power BI Desktop, you can create slicers using columns and use the slicers to filter your visual.
